AQC107: Two human semen smears stained with Eosin-Nigrosin stain
When sperm aren’t moving, the question is are they dead or do they have a motility defect? The easiest method to differentiate between the two is a viability stain based on cell membrane exclusion. We recommend eosin staining with nigrosin counterstain. The extra nigrosin step takes a few seconds and makes the eosin staining much easier to detect. Live cells exclude the stain and appear white against the dark black-purple of the nigrosin. Live sperm are stained pink. Since this is a quantitative assay, it is high complexity and needs appropriate quality control. Document the QC quickly with Levey-Jennings plots included with the product.
